Impact of co-processed excipient particles solidity and circularityon critical quality attributes of orodispersible minitablets

Mr. Arkadiusz Hejduk et. al published a paper about orodispersible minitablets (ODMTs) with co-processed excipients (CPEs).
This study investigated a group of twelve CPEs that are promoted individually in the market from the aspect of powder morphology. After that, ODMT with CPEs and coarse grade melatonin and micronized melatonin were evaluated. In the conclution, the combination of micronized melatonin and GRANFILLER-D GNF-D215 was the best for their case.

Title
Impact of co-processed excipient particles solidity and circularityon critical quality attributes of orodispersible minitablets

Abstract
The selection of appropriate functional co-processed excipient (CPE) is a crucial stage in orodispersibleminitablets (ODMTs) development. This paper aimed to identify the most important morphological attributesinfluencing theflowability and homogeneity of powder mixtures and minitabletsfinal quality. Our research in-cluded the use of the laser diffraction method, the microscopic automated imaging technique and aflowabilitytester. All powder parameters examined were divided into two groups: those affecting mixture homogeneityorflowability. A Pearson's correlation matrix revealed a strong relationship between both, especially in termsof solidity-circularity andconvexity-circularitypairs(r=−0.71,p= 0.010 andr=0.93,p= 0.000 respectively). An analysis of model mixtures confirmed that high solidity values of the CPE supportfinal mixture homogeneityand accordingly high circularity values support itsflowability. In conclusion, solidity and circularity were indi-cated as the most critical morphological parameters, which have to be analyzed to define the best possibleODMTs formulation.
